Peptone Yeast Extract Broth Based Media are enriched nonselective broth used for growth & identification of anaerobic bacteria based on their biochemical features. This media is prepared, dispensed, & packaged under oxygen-free conditions to prevent the formation of oxidized products prior to use.PY Broth contains hemin & vitamin K for the recovery of fastidious organisms such as Prevotella spp & Porphyromonas spp. PRAS (Pre-reduced anaerobically sterilized) biochemicals are considered the gold standard for biochemical testing & characterization of anaerobes. Adding specific biochemical, the Peptone Yeast Extract Broth Based Media can determine whether anaerobic bacteria will utilize the biochemical or if growth is stimulated or inhibited. PY was originally formulated for use in chromatographic analysis of metabolic products from the fermentation of glucose.

Tryptic Soy Agar with 0.03% Congo Red (TSA w/CR) is a non-selective, differential medium used for the cultivation of a wide variety of non-fastidious microorganisms. Congo red is utilized to stain the cells of fungi and the outer membrane of Gram-negative bacteria. Additionally, studies have indicated virulent Shigella flexneri serotype 2a, grown in the presence of congo red at 37°C, showed increased levels of three proteins with MRS of 43K, 58K, and 63K proteins) in the Sarkosyl-soluble membrane fractions. Baird-Parker (BP) Agar is a selective and differential medium used for the isolation and presumptive identification of coagulase-positive Staphylococcus from clinical specimens, foods, air, water, and other materials. Colonies of S. aureus form a characteristic black pigmentation by reducing the potassium tellurite and form clear zones due to proteolysis of the egg yolk emulsion. Selective Agar medium used for the isolation of Gram positive staphylococci and streptococci from clinical specimens. Eliner et al. added colistin nalidixic acid to Columbia Agar to suppress the growth of gram negative bacteria. The addition of 5-10% sheep red blood cells to the base demonstrates hemolytic patterns of the staphylococci and streptococci.
Formula per liter*:Casein Digest Peptone: 5.0g; Corn Starch: 1.0g; Peptic Digest of Animal Tissue: 8.0g; Sodium Chloride: 5.0g; Yeast Enriched Peptone: 10.0g; Colistine: 0.015g; Nalidixic Acid: 0.010g; Agar: 14.0g. *Grams per liter may be adjusted or formula supplemented to obtain desired performance.
Preparation: Mix 44 grams of the medium in a liter of purified water until evenly dispersed. Heat with repeated stirring and boil for one minute to dissolve completely. Distribute and autoclave at 121°C for 15 minutes.
